Deparaffinization is an essential step in the preparation of histopathological specimens, as it aims to remove paraffin prior to Hematoxylin-Eosin (HE) staining. Xylene is commonly used for this purpose; however, its use requires careful consideration due to the potential health and environmental risks associated with xylene exposure. This study aimed to analyze the use of olive oil as an alternative to xylene for the deparaffinization of ovarian cyst and breast cancer tissues. An experimental study was conducted using 54 histopathological specimens consisting of ovarian cyst and breast cancer tissues. Each tissue type was divided into three treatment groups: deparaffinization using xylene, olive oil heated to 60°C, and unheated olive oil, with nine specimens in each group. The quality of the specimens following HE staining was evaluated based on microscopic characteristics using a scoring system ranging from 0 to 2. The results showed that the xylene group produced 9 specimens (100%) with a score of 2. In the olive oil heated to 60°C group, 8 specimens (88.9%) received a score of 2 and 1 specimen (11.1%) received a score of 1, whereas the unheated olive oil group produced 4 specimens (44.4%) with a score of 2 and 5 specimens (55.6%) with a score of 1 for both tissue types. Statistical analysis showed no significant differences among the treatments (p > 0.05). Olive oil heated to 60°C has the potential to be used as an alternative to xylene in the deparaffinization of histopathological tissue specimens.
