Red roselle leaves (Hibiscus sabdariffa L.) contain various secondary metabolites and have potential as a source of bioactive compounds. This study aimed to determine the yield of dried material, identify secondary metabolites, and estimate the total chalcone content of 70% ethanol extract of red roselle leaves using UV–Vis spectrophotometry. A laboratory experimental design with qualitative and quantitative approaches was employed. Fresh leaves were processed into dried material and extracted by repeated maceration using 70% ethanol. Phytochemical screening was conducted for flavonoids, chalcones, alkaloids, tannins, saponins, and steroids. Total chalcone estimation was performed using chalcone standards at concentrations of 20–100 ppm. From 6,000 g of fresh leaves, 2,800 g of dried material was obtained, corresponding to a yield of 46.67%. The calibration curve produced the equation y = 0.009085x − 0.0073 with R² = 0.999909. Sample absorbances of 0.641 and 0.444 corresponded to concentrations of 71.36 and 49.68 ppm, respectively, in the measured solutions. The first and second macerates showed positive results for all tested secondary metabolite groups, and the first macerate exhibited a higher analytical response than the second macerate.
